Radioimmunoassay of thyroxine in unextracted serum, by a single-antibody technique.

چکیده
A radioimmunoassay procedure for thyroxine (14) is described, for which only 25 il of serum is required. The antibody, commercially available, is produced in rabbits in response to injections of a T4-albumin conjugate. Native T4-binding proteins are denatured by heating 25 .tl of serum in 0.2 ml of a glutamate buffer in a boiling water bath. After a 1-h incubation of heat-treated serum with l125T4 and 14 antibody, “free” and “bound” 14 are separated by use of hemoglobin-coated charcoal. The method has excellent sensitivity, is rapid, and possesses significant advantages over a standard competitive protein binding method with which it is compared.

A rapid and precise radioimmunoassay (RIA) for serum total thyroxine (T4) on as little as 1-10 mul of unextracted serum is described. Results in hypothyroidism (overt and borderline), in euthyroid subjects in pregnant and oestrogen-medicated subjects, and in hyperthyroidism (overt and borderline) are compared with the results on the same sera by an established competitive protein binding techni...
متن کاملRadioimmunoassay of triiodothyronine in unextracted human serum.
Serum triiodothyronine (T-3) concentrations have been estimated by radioimmunoassay using unextracted serum. The serum T-3 concentrations have been shown to be similar in two separate European populations (0.76-1.67 ng/ml). Raised T-3 values have been observed in all subjects with hyperthyroidism. Low values are seen in hypothyroidism although there is some overlap with the normal range. There ...

Serum triiodothyronine (T(3)) has been measured by radioimmunoassay and corroborated by analysis of the identical samples with a previously described gas-liquid chromatographic technique. Special features of the radioimmunoassay procedure which permit determinations in unextracted serum include the use of a T(3)-free serum preparation for the construction of the standard curve and of tetrachlor...
متن کاملA direct radioimmunoassay for aldosterone in unextracted serum and plasma.
We describe a novel radioimmunoassay procedure for the direct estimation of aldosterone in unextracted plasma and serum samples, in which interfering binding proteins are digested by Proteinase K (Tritirachium alkaline proteinase, EC 3.4.21.14), a powerful proteolytic enzyme. Heating at 75 degrees C for 15 min inactivates the enzyme before radioimmunoassay. Alternatively, EDTA may be used to in...
متن کاملClinical problems with a free thyroxine assay.
THE measurement of serum free thyroxine (FT4), which forms a small fraction (0.01-0.05 per cent) of the circulating total thyroxine (TT4), might be expected to provide the most accurate single biochemical index of thyroid status, on the basis that only free hormone is bioavailable. The conventional estimation of FT4 by equilibrium dialysis' is time consuming and the arrival of radioimmunoassay ...
